The direct journey from Prague to Vienna (and vice versa) takes around 4 hours which is similar to a train and a bit faster than a bus.
Prague - Vienna in one day
Depart in the morning make some sightseeing stops on the way and arrive in the evening to Vienna. Or go directly at any time - it is easily doable within one day.
Prague - Vienna with an overnight stay
Or enjoy the journey in a more relaxing way: make stops on the way and stay overnight in Český Krumlov, Znojmo, Mikulov, Brno, or any other interesting city on the way.
Sightseeing along the way
Make the most of your journey and turn an ordinary transfer into an exciting sightseeing tour! Add any interesting stop of your choice. Depending on the extra mileage, driving and waiting time, each stop is subject to additional fee which does not include the admission fees.
The route via Brno
This route is the main one connecting the two biggest Czech cities and Vienna, mostly uses highways, it allows you to stop in Brno or in southern Moravia (Mikulov or Lednice-Valtice Area).
Kutná Hora (UNESCO)
Thanks to silver mining, it used to be one of the most important Bohemian royal towns in the Middle Ages. Visit the Cathedral of St. Barbara and the Sedlec Ossuary - a place which is morbid and fascinating at the same time. Read more about Kutná Hora.

Brno

The route via Znojmo
This route is the most direct, it uses highways and lower-level inter-city roads equally. You can visit the lovely cities Třebíč, Telč, and Znojmo on the way.
Telč (UNESCO)
Telč is one of the Czech fairy-tale towns, the old town is located on a "peninsula" surrounded by fish ponds. A must-see is the square filled with colorful houses - a great example of the Italian Renaissance. Don't miss the chateau too.

Třebíč (UNESCO)
Local highlights are the UNESCO-listed Basilica of St. Procopius located in the area of the former Benedictine monastery, the Jewish Quarter, and the Jewish Cemetery.

Recommended: Znojmo
"Znojmo, vidím tě dvojmo!"

The route via Český Krumlov
This is the longest option but you can see a lot. Only roughly a third of the route uses highways the rest is a mix of all level roads including tiny regional roads which makes this route very scenic.
Hluboká Castle
On the site of the original gothic castle stands today one of the most beautiful castles in the Czech Republic, reconstructed in the romantic style of England's Windsor Castle, surrounded by English park. Admire the spectacular interiors and exteriors.
